Abstract. Inflammation, as a typical pathological process, is the basis of many diseases. With the modern approach to the treatment of inflammatory diseases, cellular technologies are used. Multipotent mesenchymal stromal cells (MMSCs), having immunomodulatory properties, can be used to reduce inflammatory processes and restore the functional state of the liver in case of toxic damage. The aim of the study was to study the effect of MMSCs on changes in the morphofunctional state of the liver during its toxic damage. Materials and methods. In the study, MMSCs were transplanted in the amount of 4 million cells/kg of weight to mice under physiological conditions and under conditions of toxic liver damage. Toxic liver damage was modeled by introducing carbon tetrachloride (ССl₄) 50 µl. per mouse intraperitoneally, once. Assessment of liver morphometric parameters was performed on day 7 after modeling of liver pathology. The activity of DNA repair enzymes of the Poly-ADP-ribosopolymerase (PARP) family was also analyzed. Results. Transplantation of MMSCs to animals with toxic liver damage leads to an increase in the mitotic activity of hepatocytes, an increase in the number of binuclear hepatocytes, and an increase in the area of the nucleus. Also, the introduction of MMSC promotes the activation of DNA repair enzymes of the PARP family, which led to a decrease in programmed cell death and a decrease in the number of hepatocytes with micronuclei. Conclusion. In this study, it was shown that MMSC transplantation does not affect the physiological regeneration of the liver, but in case of toxic damage to the liver, it provides restoration of its morphofunctional state.
