Abstract.

In the current study we investigated new preservation method that based on the xenon clathrates formation for short-term storage of the human skin grafts. For the experiments was used specially designed equipment. The sealed device is equipped with: pressure and temperature sensors to control the parameters of clathrate formation, special window enable register clathrate point in the system and the stability of this parameter over time. The skin samples were preserved in clathrates for 4 days. The damaging effect of xenon clathrates was evaluated on the stages of clathrate formation and storage by optical microscopy. The viability of preserved skin samples in xenon clathrates was evaluated by the expansion of the dermal fibroblasts in DMEM with 10 % fetal bovine serum. Histology of experimental and intact skin samples was performed by using standard hematoxilyn eosin sections. Preserved in xenon clathrates skin samples did not differ from intact specimens practically. The skin samples were stored in clathrates Xe showed a steady output of fibroblasts with positive dynamics and without difference from the intact specimens at 25 days of culture. Xenon clathrates can be used as a preservative agent for the storage of the human skin. Stages of conservation, storage and re-activation of virtually were not impact on the skin samples except some cytosol vacualization. This may be due to the re-activation process (decompression) and requires the new right findings settings in this area.
