Abstract. Introduction. Metabolic syndrome is a serious medical and social problem characterized by insulin resistance, dyslipidemia, and visceral obesity. High-fat and high-carbohydrate diets are common models for inducing metabolic syndrome in rodents. Sulodexide, a glycosaminoglycan with antithrombotic, antiatherogenic, and antioxidant effects, is considered a promising treatment for correcting metabolic disorders. Objective to evaluate the effect of sulodexide on biochemical parameters and glycosaminoglycan metabolism in rats on a high-fructose diet. Materials and Methods. The study was conducted on white nonlinear male rats for 35 days. Rodents in the control group received a standard vivarium diet at a rate of 74.4 kcal/day per rat. The first experimental group of animals, kept on a fructose diet, received 166 kcal/day per rat. The second group of rodents, kept on a fructose diet, received daily subcutaneous injections of sulodexide at a dose of 8.5 LE/kg of body weight. Then, the experimental groups received a standard vivarium diet for 25 days. Carbohydrate and lipid metabolism parameters were assessed in blood serum on days 21, 35, and 60 of the experiment; the content of glycosaminoglycans in blood, liver tissue, and skin was determined. Results. Animals on the fructose diet demonstrated hyperglycemia, hyperinsulinemia, and dyslipidemia, with changes persisting after diet discontinuation, development of insulin resistance, and a predominance of atherogenic lipoproteins. Increased glycosaminoglycans in the serum and tissues indicate the prevalence of both catabolic and anabolic processes. Administration of sulodexide during the fructose diet reduced the increase in carbohydrate and lipid metabolism parameters, as well as glycosaminoglycans in both the blood and tissues. Conclusion. A high-fructose diet induces metabolic disturbances in carbohydrate, lipid, and glycosaminoglycan metabolism in rats. Sulodexide has a positive effect on blood biochemistry and normalizes tissue glycosaminoglycan levels, indicating its potential therapeutic efficacy in the correction of metabolic syndrome.
